Its a 65U. I will be going to premix. Should I break in with 32:1 and after that go with 40:1?

I'd just go 40 to 1 all together IMHO done it for 2 65u's with great results
I coat the crank beatings and cyl's with 2 stroke oil when rebuilding so I know the first breath isn't going to get any friction

32:1 and the Yamalube semi-synthetic works great for break in.

It's harder to get a good break in with synthetic because it's such damn good oil. You need to "build some heat" to seat the rings properly and so you need to break it in much more aggressively with synthetic oil. I've had excellent luck breaking in using the yamalube 2W semi-synthetic.
